The RNA Technology Platform and Gene Manipulation Core (formerly RNAi Core) is the only resource center in Taiwan that combines "gene manipulation technology" and "recombinant viral vectors." This core facility integrates various gene manipulation reagents (RNAi, miRNA, CRISPR/Cas and cDNA) and several recombinant viral vector systems (lentivirus, retrovirus and AAV), and establishes useful service platforms to meet the needs of academic researches and support the R&D for biotech companies by providing gene manipulation-related services. The C6 core facility is one of the major CRISPR/Cas technology centers in Taiwan and can provide customized CRISPR/Cas services and assist in generating gene knockout or knock-in cell lines. The combination of CRISPR/Cas technology and recombinant AAV vectors would serve as an important tool in emerging gene therapy. Using peptide-display gene engineering, the C6 core facility has established diverse capsid libraries of AAV, which in combination with in vivo screening (e.g., in mice) can identify novel AAV vectors with higher tissue specificity. With over ten years of experience in viral preparation (lentivirus, retrovirus and AAV), the C6 core facility can also provide technical supports for the development of cell therapy. The use of optimized viral envelope protein greatly improves the transduction efficiency of recombinant lentivirus or retrovirus for immune cells. In addition to conventional lentiviral or retroviral vectors used in cell therapy, the C6 core can assist users in establishing novel CAR gene delivery system by combining AAV vector with gene manipulation tools (such as RNAi, miRNA, CRISPR/Cas).
